Acetic acid-induced visceral pain model in rat ▼

Case Study - Acetic acid-induced visceral pain model in rat & Endpoint Measurements

The acetic acid-induced writhing model is a classical and highly sensitive model for the evaluation of visceral pain and analgesic efficacy. Intraperitoneal administration of dilute acetic acid induces a characteristic writhing response, consisting of abdominal constriction, stretching of the torso, and extension of the hind limbs. These behaviors are driven by the release of inflammatory mediators and activation of visceral nociceptive pathways, making the model particularly suitable for assessing analgesic activity.
PharmaLegacy has established a robust and reproducible visceral pain platform based on quantitative assessment of writhing behaviors. The model is highly sensitive to both centrally and peripherally acting analgesics, enabling rapid evaluation of compound efficacy across a broad range of therapeutic modalities.
The predictive validity of the model has been confirmed using clinically relevant analgesics, including acetaminophen, ibuprofen, pregabalin, and opioid receptor modulators. Significant reductions in writhing behavior following treatment demonstrate the utility of the model for identifying and characterizing novel pain therapeutics.
In addition to efficacy screening, the acetic acid-induced visceral pain model can be incorporated into broader pain pharmacology programs to investigate mechanisms of nociception and analgesia. This platform is widely applicable for preclinical development of therapies targeting visceral pain, abdominal pain syndromes, inflammatory pain, and other acute pain conditions.
